WEE1‑IN‑3(Cat No.:I029900)is a potent, selective small‑molecule inhibitor of the serine/threonine kinase Wee1, a key regulator of the G₂/M checkpoint and promoter of DNA damage repair. It inhibits Wee1 with an IC₅₀ below 10 nM, causing loss of inhibitory phosphorylation on CDK1 and forcing premature mitotic entry. In vitro, it suppresses proliferation of cancer cell lines, notably SW480 (100–1,000 nM IC₅₀) and NCI‑H23 (~42 nM) . By disrupting cell cycle control, WEE1‑IN‑3 induces mitotic catastrophe and apoptosis, making it a valuable tool for studying checkpoint dependency in p53‑ or RAS‑mutant cancers.
