TMDJ-035(CAT: I040607) is a highly potent and selective inhibitor of ryanodine receptor 2 (RyR2), a calcium release channel predominantly expressed in cardiac muscle cells. With an EC₅₀ of 13 nM, TMDJ-035 effectively suppresses abnormal Ca²⁺ waves and transients in cardiomyocytes harboring RyR2 mutations, without affecting RyR1 or RyR3 isoforms . Structurally, it favors a cis-amide configuration, which is critical for its inhibitory activity .
