SHR0302 (CAT: I015377) is a potent and orally active inhibitor of the Janus kinase (JAK) family, with a particular affinity for JAK1. It exhibits selectivity for JAK1 over JAK2, JAK3, and Tyk2. By inhibiting JAK1-STAT3 phosphorylation, SHR0302 exerts anti-proliferative and anti-inflammatory effects. In hepatic stellate cells (HSCs), SHR0302 inhibits their proliferation, activation, and migration in a concentration-dependent manner. It induces apoptosis in HSCs by activating caspase-3 and Bax while reducing the expression of Bcl-2. SHR0302 also inhibits the Akt signaling pathway. In animal models, SHR0302 demonstrates efficacy in attenuating the severity of experimental arthritis and reducing inflammatory markers.
