Ro 64-6198(CAT: I009171) is a potent, selective, nonpeptide N/OFQ receptor (NOP) agonist with an EC₅₀ of 25.6 nM, exhibiting over 100-fold selectivity for NOP compared to classical opioid receptors. It demonstrates high cellular permeability and brain penetration, making it valuable for in vivo studies. Ro 64-6198 produces functional desensitization of NOP receptors and recruits arrestin3 (EC₅₀ = 0.912 μM) and arrestin2 (EC₅₀ = 1.20 μM) in a concentration-dependent manner. Preclinically, it shows anxiolytic effects in neophobia-based assays, including marble burying, elevated plus maze, and open field tests, without impairing locomotion. Ro 64-6198 is widely studied in anxiety, addiction, neuropathic pain, cough, and anorexia research.
