L-745870 hydrochloride(CAT: I017523) is a potent and highly selective dopamine D4 receptor antagonist (Ki = 0.43 nM) with excellent brain penetration and oral bioavailability. It exhibits substantially weaker affinity for D2 and D3 receptors, and moderate activity at 5-HT2 receptors, sigma sites, and α-adrenoceptors. Functionally, L-745870 blocks D4 receptor–mediated inhibition of adenylate cyclase activity, calcium currents, and G protein–coupled inwardly rectifying K⁺ channels, as well as extracellular acidification responses in transfected cells. Pharmacokinetic studies show favorable half-life (2.1–2.8 h) and high brain-to-plasma ratios. In vivo, it induces mild sedation at higher doses, supporting its utility in dopaminergic research.
