CINPA1(CAT: I011252) is a potent and selective constitutive androstane receptor (CAR) inhibitor that does not activate the pregnane X receptor (PXR), making it a valuable molecular probe for studying xenobiotic receptor signaling. It reduces CAR-mediated transcription with an IC50 of ~70 nM and blocks CAR transactivation in HepG2 cells without PXR activation. In primary human hepatocytes, CINPA1 suppresses CAR-dependent gene expression without altering CAR protein levels or localization. Mechanistically, CINPA1 increases corepressor binding, reduces coactivator recruitment, and disrupts CAR-promoter interactions, confirming its role as a CAR ligand. It is widely applied in drug metabolism, toxicology, and liver disease research.
