ZK53(CAT: I040304) is a selective small-molecule activator of human mitochondrial caseinolytic protease P (HsClpP), exhibiting an EC₅₀ of 1.37 μM for α-casein hydrolysis. Unlike other ClpP modulators, ZK53 is inactive against bacterial ClpP, highlighting its human-specific selectivity. By activating HsClpP, ZK53 induces mitochondrial dysfunction and apoptosis in lung squamous cell carcinoma (LUSC) cell lines such as H1703, H520, and SK-MES-1. It also impairs mitochondrial homeostasis and bioenergetics, ultimately suppressing tumor growth in H1703 xenograft mouse models. ZK53 is a valuable research tool for investigating mitochondrial proteostasis and ClpP-related mechanisms in cancer.
