W1131 TFA(CAT: I040301) is a potent inhibitor of STAT3 signaling that induces ferroptosis and exhibits strong anticancer efficacy. It significantly suppresses tumor progression in gastric cancer models, including subcutaneous xenografts, organoids, and patient-derived xenografts (PDX). W1131 enhances sensitivity to 5-fluorouracil (5-FU, HY-90006), effectively overcoming chemoresistance. Mechanistically, it regulates multiple cellular pathways, including the IL6-JAK-STAT3 axis, ferroptosis, cell cycle progression, DNA damage response, and oxidative phosphorylation. As a trifluoroacetate salt, W1131 TFA offers improved solubility and stability for in vivo and in vitro applications. This compound is a valuable research tool for exploring targeted therapies in gastric and other STAT3-driven cancers.
