Val-Cit-PAB-OSBT(CAT: I040448) is a cleavable linker used in the synthesis of antibody-drug conjugates (ADCs), engineered to enable selective drug release within target cells. It consists of the dipeptide Valine-Citrulline (Val-Cit), a self-immolative para-aminobenzyl (PAB) spacer, and a reactive OSBT (ortho-succinimidyl benzoyl thioester) group that facilitates stable conjugation to payloads or antibodies. Upon internalization into lysosomes, the Val-Cit linker is enzymatically cleaved by cathepsin B, triggering PAB-mediated release of the cytotoxic drug. This modular structure supports efficient and controlled drug delivery, making Val-Cit-PAB-OSBT a key component in the development of targeted, tumor-selective therapeutic ADCs with enhanced safety profiles.
