TMP920(CAT: I017430) is a highly potent and selective RORγt antagonist, inhibiting RORγt binding to the SRC1 peptide with an IC50 of 0.03 μM. In cellular studies, TMP920 (0.09–30 μM) suppresses Th17 cell differentiation and IL-17 production, while displaying cytotoxicity only at concentrations >10 μM in naïve CD4⁺ T cells. At submicromolar levels (<2.5 μM), TMP920 effectively blocks IL-17 secretion and downregulates the Th17 transcriptome, while promoting alternative T-cell subsets. Mechanistically, it reduces RORγt-DNA interactions, making it a valuable tool compound for investigating autoimmune disease mechanisms, inflammation, and T-cell biology through modulation of Th17 signaling pathways.
