TMP778(CAT: I046023) is a potent and selective inverse agonist of RORγt, with an IC₅₀ of 7 nM in FRET assays. It efficiently suppresses IL-17 production by inhibiting Th17 cell differentiation and activity without affecting RORγt expression, nuclear translocation, or general cell proliferation at non-toxic concentrations. In vivo, TMP778 treatment delays disease onset and markedly reduces severity in experimental autoimmune encephalomyelitis (EAE) models. It decreases CNS infiltration by mononuclear cells and significantly reduces IL-17⁺ T cells, while sparing Th1 responses. With strong efficacy against Th17-mediated inflammation, TMP778 is a valuable tool for autoimmune disease research and immunomodulatory drug development.
