TD-0212 TFA(CAT: I017437) is an orally active dual-acting small molecule that functions as both an angiotensin II type 1 receptor (AT1) antagonist (pKi = 8.9) and a neprilysin (NEP) inhibitor (pIC₅₀ = 9.2). By combining AT1 blockade with NEP inhibition, TD-0212 enhances antihypertensive efficacy while potentially lowering the risk of angioedema compared with dual ACE/NEP inhibition. In preclinical hypertension models, TD-0212 demonstrates blood pressure reductions comparable to omapatrilat and to combinations of AT1 receptor antagonists with NEP inhibitors. This pharmacological profile makes TD-0212 TFA a valuable research tool for cardiovascular disease, hypertension, and renin–angiotensin–neprilysin system modulation.
