SK33(CAT: I016844) is a trifluoromethylated enobosarm analog that acts as a potent, tissue-selective anti-androgen by reducing androgen receptor (AR) transcriptional activity. In prostate cancer models, SK33 demonstrates strong inhibitory effects with an IC₅₀ of 0.2 μM in LNCaP-AR⁺ cells, where it significantly reduces S-phase entry. In LNCaP/BicR cells, SK33 suppresses cell growth in a dose-dependent manner with an IC₅₀ of ~5 μM after 96 hours. In vivo, SK33 (50 mg/kg; s.c.; 24 h) effectively inhibits AR-driven transcription. With its potency and tissue selectivity, SK33 represents a promising probe for AR-targeted cancer research and therapeutic exploration.
