R 59-022 (CAT: R024299) is a diacylglycerol kinase (DGK) inhibitor with an IC₅₀ of 2.8 μM that blocks the phosphorylation of OAG to OAPA. Beyond DGK inhibition, it functions as a 5-HT receptor antagonist and activator of protein kinase C (PKC). In vitro, R 59-022 potentiates thrombin-induced platelet aggregation, enhances diacylglycerol production, and inhibits phosphatidic acid formation in neutrophils. It also stimulates noradrenaline release in chromaffin cells and activates PKC in tumor cell lines. Notably, R 59-022 blocks Ebola virus entry in Vero cells and extends survival in glioblastoma-bearing SCID mice, making it a versatile tool for cancer, virology, and signaling research.
