PS10(CAT: I016426) is a novel, potent, ATP-competitive pan-PDK inhibitor with activity against all pyruvate dehydrogenase kinase isoforms. It demonstrates IC₅₀ values of 0.8 μM (PDK2), 0.76 μM (PDK4), 2.1 μM (PDK1), and 21.3 μM (PDK3), showing strongest affinity toward PDK2 (K_d = 239 nM) compared to Hsp90 (K_d = 47 μM). By inhibiting PDK activity, PS10 enhances pyruvate dehydrogenase function, promotes myocardial carbohydrate oxidation, and improves glucose tolerance in models of diet-induced obesity. This compound is a valuable tool for investigating metabolic regulation and holds therapeutic potential in diabetic cardiomyopathy and other metabolic disease research.
