Pozanicline dihydrochloride (CAT: R036177) is an orally bioavailable and selective nicotinic acetylcholine receptor (nAChR) agonist with high affinity for α4β2 receptors (Ki = 16.7 nM at [³H]cytisine sites). It binds rat brain α4β2 nAChRs with a Ki of 17 nM, showing minimal activity at α7 receptors. As a partial agonist, Pozanicline exhibits high selectivity for α6β2 and α4α5β2 subtypes, with notable potency at α6β2 (EC50 = 0.11 μM). In vivo, Pozanicline (0.3–0.6 mg/kg) reverses nicotine withdrawal-induced cognitive deficits, highlighting its utility in neuropharmacology, cognition, addiction, and neurological disease research.
