PF-04995274(CAT: I008671) is a potent, high-affinity, and orally active partial serotonin 4 receptor (5-HT4R) agonist with nanomolar activity across multiple isoforms. It shows EC50 values of 0.26–0.47 nM for human 5-HT4A/4B/4D/4E (Ki 0.15–0.46 nM) and 0.59–0.65 nM for rat 5-HT4S/4L/4E (Ki 0.30 nM for rat 5-HT4S). Brain-penetrant and pharmacologically robust, PF-04995274 has demonstrated efficacy in cognitive and behavioral models. In mice, repeated administration (3–10 mg/kg, i.v., 17 days) attenuates learned fear and reduces stress-induced depressive-like behaviors. With its strong receptor selectivity and CNS penetration, PF-04995274 is a valuable research tool for Alzheimer’s disease and neuropsychiatric disorders.
