MS31 trihydrochloride(CAT: I045993) is a potent, high-affinity, and selective inhibitor of the methyllysine reader protein spindlin 1 (SPIN1). It effectively blocks SPIN1 interaction with histone H3K4me3, showing inhibitory activity with IC₅₀ values of 77 nM (AlphaLISA) and 243 nM (FP). MS31 binds specifically to the Tudor domain II of SPIN1 (Kd = 91 nM), preventing recognition of trimethyllysine-containing peptides. With strong selectivity over other epigenetic readers and writers, it directly engages SPIN1 in cells while demonstrating minimal toxicity in non-tumorigenic cells. MS31 trihydrochloride is a valuable chemical probe for studying SPIN1-dependent transcriptional regulation and epigenetic signaling.
