MMH2(CAT: I040274) is a novel BRD4 molecular glue degrader that selectively targets the second bromodomain of BRD4 (BRD4^BD2). Unlike traditional PROTACs, MMH2 functions by stabilizing the interaction between BRD4 and the E3 ligase complex CUL4-DCAF16, promoting ubiquitination and proteasomal degradation of BRD4. This mechanism offers a unique approach to epigenetic regulation by eliminating BRD4, a key transcriptional coactivator implicated in cancer and inflammation. MMH2 is a valuable chemical probe for studying targeted protein degradation via molecular glue strategies and BRD4-mediated transcriptional control.
