ML-180 (CAT: R065005) is a potent inverse agonist of the orphan nuclear receptor liver receptor homolog 1 (LRH-1; NR5A2) with an IC₅₀ of 3.7 µM, showing inactivity toward steroidogenic factor-1 (SF-1; NR5A1; IC₅₀ >10 µM). In hepatic cells, ML-180 inhibits LRH-1 signaling, suppresses cyclin-D1/E1 expression, and reduces LRH-1 mRNA and downstream targets (CYP19, GATA3, GATA4) in a dose-dependent manner. It efficiently blocks proliferation in LRH-1-positive Huh-7 cells (EC₅₀ ≈ 2.8 µM). In vivo, ML-180 (30 mg/kg, i.p.) decreases LRH-1 and SHP mRNA in adrenal and pancreatic tissues, supporting its potential in LRH-1-dependent cancer research.
