KY-226(CAT: I017426) is a potent, selective, orally active, allosteric inhibitor of protein tyrosine phosphatase 1B (PTP1B) with an IC₅₀ of 0.25 μM and no PPARγ agonist activity. By enhancing insulin and leptin signaling, KY-226 demonstrates strong anti-diabetic and anti-obesity effects. In vitro, it increases insulin-stimulated phosphorylation of insulin receptors in HepG2 cells and restores Akt/FoxO1 signaling and tight junction protein ZO-1 under inflammatory stress. In vivo, oral administration reduces plasma glucose, triglycerides, and HbA1c levels in db/db mice without affecting weight gain. KY-226 also protects neurons against cerebral ischemic injury, supporting its potential in metabolic and neurological disease research.
