KIRA-7(CAT: I017148) is an imidazopyrazine compound that binds the IRE1α kinase domain and allosterically inhibits its RNase activity (IC₅₀ = 110 nM). By suppressing IRE1α-mediated signaling, KIRA-7 effectively reduces XBP1 splicing and downstream unfolded protein response (UPR) activation. In alveolar epithelial MLE12 cells, it inhibits XBP1 splicing, while in C57BL/6 mice (5 mg/kg, i.p., 14 days), KIRA-7 significantly decreases spliced XBP1, ATF4, BiP, and CHOP levels after bleomycin exposure. Additionally, it lowers expression of fibrosis-associated genes, including collagen 1A1 and fibronectin, demonstrating strong anti-fibrotic potential. KIRA-7 is a valuable probe for studying ER stress, UPR signaling, and fibrosis.
