JH-LPH-28(CAT:I016924) is a sulfonyl piperazine analog and potent inhibitor of UDP-2,3-diacylglucosamine pyrophosphate hydrolase (LpxH), a key enzyme in the lipid A biosynthesis pathway of Gram-negative bacteria. It exhibits strong enzymatic inhibition with IC₅₀ values of 0.11 μM against Klebsiella pneumoniae LpxH and 0.083 μM against Escherichia coli LpxH. JH-LPH-28 demonstrates outstanding antibacterial activity, with a MIC of 0.83 μg/mL, and effectively inhibits wild-type K. pneumoniae growth with a MIC of 1.6 μg/mL. By targeting LpxH, JH-LPH-28 represents a promising lead compound for antibiotic development against multidrug-resistant Gram-negative pathogens.
