ICA-27243(CAT: I007250) is a selective, potent, and orally active KCNQ2/Q3 potassium channel opener with an EC50 of 0.38 μM, showing reduced activity at KCNQ4 and KCNQ3/Q5. In SH-SY5Y neuroblastoma cells, it induces membrane hyperpolarization, reversible by M-current inhibitors XE-991 and linopirdine. ICA-27243 enhances 86Rb⁺ efflux (EC50 = 0.2 μM) and increases whole-cell currents in CHO cells expressing KCNQ2/Q3 channels (EC50 = 0.4 μM), producing a hyperpolarizing V1/2 shift of –19 mV at 10 μM. Orally administered in mice, ICA-27243 displays anticonvulsant activity in the maximal electroshock model (ED50 = 8.4 mg/kg), supporting its potential in epilepsy research.
