HPN-01(CAT: I017321) is a potent and selective IκB kinase (IKK) inhibitor, showing pIC50 values of 6.4 for IKK-α, 7.0 for IKK-β, and <4.8 for IKK-ε, with >50-fold selectivity over a broad kinase panel. It effectively suppresses LPS-induced pro-inflammatory cytokine secretion (TNF-α, IL-1β, IL-6) in human PBMCs and blocks TNF-α–driven NF-κB nuclear translocation in lung fibroblasts. Beyond its immunomodulatory effects, HPN-01 inhibits SREBP-1/2 expression in hepatocytes (IC50 = 1.71 μM, 3.43 μM), linking it to lipid metabolism regulation. With dual anti-inflammatory and metabolic actions, HPN-01 is being developed as a therapeutic candidate for nonalcoholic fatty liver disease (NAFLD).
