Gut restricted-7 (CAT: I045981) is a potent, covalent, and orally active pan–bile salt hydrolase (BSH) inhibitor with selective gut restriction. It effectively reduces gut bacterial BSH activity and decreases deconjugated bile acid levels in mouse feces without altering microbial biomass. At 60 μM, GR-7 preserves epithelial barrier integrity in Caco-2 cells, demonstrating low cytotoxicity. In vivo, dietary administration (0.09% w/w) significantly suppresses fecal BSH activity within 8 hours, with ~20 μM detected in cecal contents. By targeting intestinal BSHs while remaining gut-selective, GR-7 is a valuable chemical probe for studying bile acid metabolism, host–microbiome interactions, and metabolic disease pathways.
