DTSSP (3,3′-Dithiobis[sulfosuccinimidylpropionate])(CAT: I014349) is a water-soluble, homobifunctional, cleavable crosslinker commonly used for protein–protein interaction studies. It contains two NHS ester groups that efficiently react with primary amines on lysine residues or N-termini of proteins, forming stable amide bonds. The two reactive ends are connected by a disulfide-containing spacer arm (~12 Å), which can be selectively cleaved under reducing conditions (e.g., with DTT or TCEP), allowing reversible crosslinking and downstream analysis of interacting partners. Its sulfonate groups confer excellent aqueous solubility, eliminating the need for organic solvents. DTSSP is widely used in structural biology, proteomics, and immunology for mapping protein complexes and stabilizing transient interactions.
