CB1151(CAT: I017272) is a synthetic 20-epi analogue of 1,25-dihydroxyvitamin D₃ (VD₃) with potent antitumor activity. In MCF-7 breast cancer cells, CB1151 suppresses proliferation with an IC₅₀ of 0.82 nM, as measured by [³H]-thymidine incorporation. It selectively activates vitamin D response elements, showing greater potency for IP9-type VDREs (EC₅₀ = 1.2 nM) compared to DR3-type elements (EC₅₀ = 3.2 nM), correlating strongly with its antiproliferative effects. CB1151 also exhibits a functional dissociation constant (Kdf) of 3.6 nM, reflecting its stable interaction with the vitamin D receptor (VDR).
