BIP-135(CAT: R040970) is a potent and selective ATP-competitive glycogen synthase kinase-3 (GSK-3) inhibitor, with IC50 values of 16 nM (GSK-3α) and 21 nM (GSK-3β). It demonstrates strong neuroprotective activity, enhancing survival motor neuron (SMN) protein expression in human SMA fibroblasts (25 μM, 72 h) with a bell-shaped dose-response profile. In oxidative stress models, BIP-135 (20 μM, 48 h) shows superior neuroprotection. In vivo, treatment (75 mg/kg i.p., P0–21) prolongs median survival in the Δ7 SMA knockout mouse model of spinal muscular atrophy, with no significant toxicity or weight loss, highlighting its promise in neurodegenerative disease research.
