BI-2545(CAT: I017408) is a potent autotaxin (ATX) inhibitor that effectively reduces lysophosphatidic acid (LPA) signaling. It shows strong inhibitory activity with IC50 values of 2.2 nM for human ATX and 3.4 nM for rat ATX. In functional assays, BI-2545 demonstrates potency in LPA and rat whole blood assays with IC50s of 29 nM and 96 nM, respectively. Administered orally at 10 mg/kg, BI-2545 exhibits high and sustained in vivo efficacy in lowering LPA levels, with a pharmacokinetic half-life (t1/2) of 3.4 hours. This compound is a valuable tool for exploring ATX biology and LPA-mediated disease mechanisms.
