Ac4ManNAz (80% α isomer)(CAT: I016663) is a peracetylated N-azidoacetylmannosamine derivative used as a metabolic glycoprotein labeling reagent. Once taken up by cells, it is incorporated into sialic acid residues of glycoproteins, enabling selective modification through azide-mediated bioorthogonal chemistry. The azide functionality reacts efficiently via copper-catalyzed azide–alkyne cycloaddition (CuAAC) or strain-promoted alkyne–azide cycloaddition (SPAAC) with alkyne, DBCO, or BCN groups. At 10 μM, Ac4ManNAz provides effective labeling with minimal cellular perturbation, while higher concentrations (e.g., 50 μM) can impair energy metabolism, infiltration ability, and channel activity. This reagent is widely applied in cell labeling, tracking, and proteomic analysis.
