A-841720（Cat No.:I000318）is a potent, non-competitive, and selective antagonist of the metabotropic glutamate receptor 1 (mGluR1), exhibiting an IC₅₀ of 10 nM for human mGluR1. It demonstrates 34-fold selectivity over mGluR5 (IC₅₀ = 342 nM) and shows negligible activity across various other neurotransmitter receptors, ion channels, and transporters. In preclinical studies, A-841720 has shown analgesic potential, significantly attenuating spontaneous post-operative pain behavior in rats with an ED₅₀ of 10 μmol/kg. However, at higher doses, it may induce motor and cognitive side effects, which could limit its therapeutic application.
