β-catenin

β-catenin

β-catenin consists of 781 amino acids, which are composed of amino terminal, carboxyl terminal and 12 arm motifs. There are 42 serine, 48 threonine, 17 tyrosine and 26 lysine. β-catenin is a double-functional protein that has different localization and functions in cells. It regulates cell-cell adhesion and coordinates gene transcription. On the one hand, it can be used as the core molecule of Wnt/β-catenin signal pathway, which can be expressed in the cytoplasm or nucleus under the action of Wnt signal and start the downstream events of the pathway. On the other hand, it can be expressed on the cell membrane as an adhesive molecule between cells. Under normal conditions, β-catenin, together with E-cadherin and a- catenin, binds to the cytoskeletal proteins of the cell membrane and maintains cell polarity and stability. When cell membrane β-catenin is lost, adhesion between cells decreases, and cells become loose and easy to fall off and migrate.

Inhibitor of β-catenin

I XAV-939 is a selective β-catenin-mediated transcriptional inhibitor and a stabilizer of axin, which can inhibit the activity of TNKS1 and TNKS2. IC 50 values are 5 and 2 nm respectively.

II Salinomycin is a Wnt/ shell-catenin channel inhibitor acting on Wnt/Fzd/LRP complex. Salinomycin strongly inhibited the reported gene activity of Wnt1 stimulation, while IC 50 was 163 nM, and decreased the level of β-catenin.

III FH535 is an inhibitor of Wnt/ β-catenin and PPAR and has antitumor activity.

IV KYA1797K is effective and has a selective Wnt/β-catenin inhibitor with an IC 50 value of 0.75 μM.

β-catenin and diseases

Under normal circumstances, β-catenin is mainly expressed in the cell membrane, and when it is expressed in the cytoplasm or nucleus, it is believed that the activation of Wnt/β-catenin pathway will promote the occurrence and development of various tumors. Abnormal phosphorylation of β-catenin will lead to biological dysfunction of β-catenin, which may be closely related to the occurrence of various diseases such as and diabetes . Therefore, the study of β-catenin in cell abnormal proliferation, skin cancer, nasopharyngeal cancer, liver cancer and other diseases and their phosphorylation has become a hot spot.
